1、浙江理工大學碩士論文摘要在對家蠶蛹eDNA文庫測序中,篩選到一個家蠶未知功能基因BIn-X,通過對其eDNA序列生物信息學分析。該eDNA序列全長為778bp,開放閱讀框(ORF)長444bp,其編碼的蛋白質(zhì)Bm-X,全長147個氨基酸殘基,預測相對分子量為16.4kI)a,等電點為3.69,為無信號肽的非分泌的酸性蛋白。經(jīng)PCR擴增得到Bm-X基因片段,經(jīng)BamHI和XhoI雙酶切后,插入相應酶切后質(zhì)粒pET-28a(+),構(gòu)建重組融合蛋白表達載體pET-28a(+)坦m出經(jīng)PCR、酶切鑒定重組正確。將構(gòu)建的重組質(zhì)粒轉(zhuǎn)化大腸桿菌Rosseta(DE3)菌
2、株感受態(tài)細胞,篩選重組菌株后以IPTG誘導表達,裂解菌體作SDS-PAGE分析,在20.1kD處有一濃集的特異性蛋白條帶,表達量可達菌體總蛋白的50%左右。融合蛋白以可溶形式存在,超聲波裂解菌體后離心收集上清液,采用鎳離子親和層析柱純化重組融合蛋白rBm-X。以該融合蛋白為抗原免疫雄性新西蘭兔制備了多克隆抗體,ELISA檢測該抗體血清的效價可達到1:8000以上。Westernblotting檢測表明制備的多克隆血清具有較高的特異性。隨后以多克隆抗體為工具,對Bm-X基因進行亞細胞定位,并分析Bm-X基因在五齡家蠶各組織蛋自和蛹中的表達分布:Bm.X在家蠶
